Abstract:

BACKGROUND: Stem cells have great effects in clinical cell-based therapy. Accordingly, controlling the behavior and directing the fate of stem cells cultured in the laboratory is an important issue. OBJECTIVES: The aim of this study was to evaluate osteogenic properties of adipose derived mesenchymal stem cells (ADSCs) which differentiated toward osteogenic linage by osteoblast-imprinted substrate. METHODS: Rat ADSCs seeded on osteoblast-imprinted substrates, alkaline phosphatase (ALP) were measured in cellular supernatant of days 7 and 14. Alizarin Red staining of mineral matrix production in day 17 was performed. In order for in vivo evaluation, after seeding differentiated cells on a simple collagen scaffold, samples were implanted in an 8mm critical size calvarial defect. After 4 weeks defect site was harvested and prepared for histopathological examination. RESULTS: ALP in both time points was significantly more than in undifferentiated ADSCs. (p

Rat Bone Marrow Mesenchymal Stem Cell Differentiation to Insulin Producing Cells and Evaluation their Responses in Vitro and in Vivo

Background  In recent years, many researchers haveattempted to cure diabetes by using stem cells technology. Stem cells from different sources have capabilityto differentiateinto insulin producing cells (IPCs) by different methods. The obstaclesof these methods aretheirexpensive materials and complexity ofmethodswhichare practicallydisadvantagesfor producing enough transplantableIPCs that can ...

Osteogenic Differentiation of Rat Mesenchymal Stem Cells from Adipose Tissue in Comparison with Bone Marrow Mesenchymal Stem Cells: Melatonin As a Differentiation Factor

Background: Adipose-derived stem cells (ADSC) could be an appealing alternative to bone marrow stem cells (BMSC) for engineering cell-based osteoinductive grafts. Meanwhile, prior studies have demonstrated that melatonin can stimulate osteogenic differentiation. Therefore, we assayed and compared the melatonin effect on osteogenic differentiation of BMSC with that of ADSC. Methods: Mesenchymal...

In vitro differentiation of rat mesenchymal stem cells to hepatocyte lineage

Objective(s): Mesenchyme is a type of undifferentiated loose connective tissue that is derived mostly from mesoderm. Recently, mesenchymal stem cells (MSCs), as adult stem cells (ASCs) able to divide into a variety of different cells, are of utmost importance for stem cell research. In this research, ability of the liver extract to induce differentiation of rat derived omentum tissue mesenchyma...

The potential of human-derived periodontal ligament stem cells to osteogenic differentiation: An In vitro investigation

Background: Periodontal ligament stem cells (PDLSCs) are considered as a type of mesenchymal stem cell that is beneficial target for numerous clinical applications in periodontal tissue regeneration therapy. Materials and Methods: This study examined the effects of dexamethasone (Dex) on human PDLSCs in vitro. PDLSCs obtained from the roots of patient’s teeth were cultured with Dex (0....

In-vitro Differentiation of Human Umbilical Cord Wharton’s Jelly Mesenchymal Stem Cells to Insulin-Producing Cells

Background & Objective: Diabetes is a major chronic metabolic disease in the world. Islet transplantation is a way to treat diabetes. Unfortunately, this method is restricted due to graft rejection and lack of donor islets. Mesenchymal Stem Cells (MSCS) have the ability to differentiate into Insulin-Producing Cells (IPCs). In this study, Human Umbilical Mesenchymal Stem Cells (HUMSCS) were in...
